New method - Ketone bodies analysis by GC-MS

 

Ketone bodies consist of three water-soluble compounds, namely Acetone, Acetoacetate, and 3-OH-butyrate. They are produced in the liver during starvation or carbohydrate restriction as source of energy instead of glucose. Two of them are used as a source of energy in the heart and brain, while acetone arises as a breakdown product of acetoacetic acid after decarboxylation.

We have developed a combination of two GC-MS methods for the specific and sensitive determination of acetone and acetoacetate on the one hand and 3-OH-butyrate on the other hand in plasma and brain tissue samples, respectively. The methods are validated according to FDA guidlines and are suitable for the application in routine analysis.
